Phytoestrogens are capable of acting as estrogen agonists or antagonists in mammalian estrogen-sensitive pathways. Of particular interest are the potential effects these compounds may have on adenohypophyseal activity. We investigated phytoestrogen treatment alone and in combination to assess their ability to augment estrogen response element (ERE)-mediated gene expression in pituitary adenoma cells. Following preliminary binding curve studies, rat pituitary adenoma cells (RC-4B/C) were transfected with an estrogen responsive reporter (MMTV-ERE-Luc). Cells were treated for up to 48 hours with estradiol-17beta (1nM) or one of three phytoestrogens (equol, daidzein, genistein; 0.01 to 1000 nM) alone and in combination. Whereas the individual phytoestrogen treatments did increase ERE-mediated transcription at select dosages and time points, the combinatorial treatments did not augment (p > 0.05) ERE-mediated gene activity
